After Unlocking Vlad Tepes Dracula
- Dario: Complete any stage with Charlotte Aulin, using only Raging Fire.
- Olrox: In the left room of the Dinner Hall, defeat him with Dark Rift.
- Walter: On the bridge between the front and middle buildings, defeat him as Rinaldo Gandolfi.
- Graham: Defeat all bosses in the Ode to Castlevania stage in a single run.
- Kermet: On the top floor of the Laboratory, defeat it as a team with two characters (e.g., Jonathan & Charlotte).
- Frozenshade: Turn a certain number of Frozen Shades into jewels with the Jewel Gun.
- Familiar: As Julia Laforeze, obtain two Familiar Forge items in a run. This can be done by picking one from the Candybox and then another as a stage item.
- Innocent Devil: All familiars gain +1 Amount every 20 levels (up to +3).
- Maria: Play as Maria Renard with the Gemini arcana equipped, and evolve Peachone and Ebony Wings into Vandalier.
- Cornell: As Cornell, evolve the Custos weapons and kill 100,000 enemies while transformed.
- Ferryman: With Rapidus Fio equipped, dash alongside the water area near the castle gates.
- Master Librarian: Unlock the Librarian as a merchant by acquiring the Wood Carving Stone (dropped by the Treant boss in the southwestern part of the stage map), then purchase each weapon he sells at least once.
- Hammer: Survive 30 minutes wielding six evolved weapons in the Coat Of Arms category.
- Wind: Activate Eric Lecarde’s Burning Alcarde ability at least three times in a single run.
- Brauner: With the Blood Astronomia arcana equipped, kill enough Living Painting enemies in the Doppelganger arena area after 24 minutes.
- Soleil Belmont: Defeat the secret boss Soleil Belmont while using Sanctuary. The boss is located on the first tower of the castle, between the second and third teleporters.
- Dmitri Blinov: Kill Menace, the giant zombie boss in the Sewers, while playing as Celia Fortner.
- Celia Fortner: Evolve Umbra and Luminatio, then max their levels. Open a chest to evolve them into Universitas, their final form.
- Graham Jones: Defeat every boss in the Ode To Castlevania stage in a single run with any character (Endless Mode advised).
- Carmilla: Kill Carmilla, located to the right of the King’s Gate teleporter, with a Nightmare spellbook.
- Cave Troll: Defeat enough Cave Trolls across multiple runs.
- Fleaman: Defeat enough Fleamen across multiple runs.
- Axe Armor: Kill enough Axe Armors using the Stellar Blade.
- Succubus: Kill the Succubus boss using the Nightmare spellbook.
- Alamaric Sniper: Kill enough Alamaric Snipers using the Stellar Blade.
- Blackmore: As Barlowe, kill the Blackmore boss in the hidden Library area near the coffin where Reinhardt is unlocked.
- Malphas: As Quincy or John Morris, kill the Malphas boss (an angel with black wings) that spawns after the 25-minute mark.
- Death: As Richter Belmont, get the Clock Tower weapon, then go to the throne room for a rematch.
- Loretta & Stella: In the tower before the Throne Room, defeat Loretta and Stella with the Sanctuary spellbook equipped.
After unlocking Serpent Gate:
- Joachim: In the right room of the Crystal Cave, defeat him as Sara Trantoul.
After unlocking Beast Gate:
- Galamoth: Located in the center of the maze, defeat it with Power of Sire.
- Chaos: Unlock every secret in the DLC.
